Do we need to have very deep knowledge of Java for automation testing?

No, you don't need very indepth knowledge of java for automation testing. Only basics like variables/classes/conditions and loops & OOPS concept is enough to start with automation testing.

What is the best automation test tool?

Tricentis Tosca Automation tool as it is easy to learn and use when compared to other automation tool like Selenium with java etc. Only concern with the tool would be about pricing/purchasing the tool license.
